Last 2 weeks, I presented Tilly’s recent (Feb 2012) research article on isolation of oogonial stem cells from healthy reproductive-age mice and also women during my stem cell themed presentation in Special Topics in Human Genetics.

This issue has been a very very controversial and exciting since year 2004 when Tilly discovered and named the oogonial stem cells in mammary ovaries. In 2005, they showed bone marrow transplantation rescues fertility in female mice that exposed to sterilising doses of chemotherapy. This brings to a hypothesis which says oocytes may derived from circulating bone marrow cells which migrate to the ovaries. The next year, Wagers and colleagues disproved this hypothesis by using parabiotic mouse models. Now they come to this level of work where the whole isolation protocol is fully established, however, many people still doubt the reliability of OSCs.

Frankly, it never cross my mind to question this research article because Tilly and colleagues done all the necessary test to confirm OSCs are indeed stem cell and capable of forming oocytes in the mouse. Not until I found this and this through Google+.

Discovery of OSCs itself is already a kind of disproval on our centre dogma of reproduction. However, doubts are keep surrounding the GFP detected in oocytes, the occytes itself and also the function of OSCs in human ovaries since female reproduction is time-limited.

Their comments really struck my mind and open mysepeteyes. Yes, duck. I never thought of asking duck to prove them as a duck by giving birth to some ducklings. I thought stem cell markers & oocyte markers expression, tumour formation and differentiation into oocytes are sufficient to confirm OSCs are truly OSCs…..

Science is so amazing and challenging, everything is within the rules but yet still not bounded by the rules. Cheers! sure a long way to go for me =D

My 1st self-made 6x DNA loading dye by using glycerol~! Amazingly dark blue in colour and heavier compared to the commercial one. Once we load it, it will sink right into the bottom of the well as compared to the commercial one, but diffuse faster compared to those made with Ficoll-400, thus yielding a rather not-so-sharp band.

Recipe?

For 10ml of 6x loading dye,

6ml of glycerol (final conc. = 60% w/v)

1ml of 1M Tris-HCl, pH 7.4 (final conc. = 100mM Tris-HCl)

2ml of 0.5M EDTA (final conc. = 100mM EDTA)

0.025g / 25mg bromophenol blue powder

then, top up to 10ml with Millipore water.

Busy rescuing my “My PhD” “My video” “AVG Games” “All novel” bla bla folders by transferring all of them into Dr. M’s NBGG Backup external hard disk. Munfun & Jiapei also offer their external or pendrive but I’ve got 115 cloud!

Hard disk and 115 cloud, double insurance. *thumbs up*

After messing with it for several hours, suddenly it asked me “want to repair?”, SURE!

Done, it’s recovered but for god sake, i’m going to certified death on it. I changed its brain for 3 times since 2007!! Well, it’s time for a new toy.

Questions like “Where i can get a credit card for installment?!” “Should i get myself a credit card with some agreement with my bosses (for cheating)?” “Which one i should buy?”

Anyway, it still can tahan for several months i guess, no rushing as long as everything is back up-ed everyday. Yeap, everyday.

As i discuss with Panda about 妖孽 and Max Barskih，she recommended me another Ukrainian dancer called “Kazaky”.

Seriously, they are smoking hot and sexy! I like the way they present the dance in a such feminine but yet so masculine and energetic way. It somehow reminds me of the dance part in dance competition such as Vibe Dance Competition and also my first love - CADC!!!

I am seriously in love with this video and him, Max Barskih, 他TMD真是个妖孽。
